The A.I. is using Racing Hard tires, so that's the primary thing. Your objective shouldn't be to smoke the A.I., but merely to beat them. In dry conditions, they'll be doing anywhere between 3:30 and 3:50, depending on their aggressiveness, so ideally you'll want to be able to do 3:30s on Racing Hards or Mediums easily and consistently.
As for settings, you'll want to use a high-downforce setup. The speed lost on the backstraight is more than made up with the speed gained through the twisty front-sections. You might also consider using a slightly longer gearbox than you would think necessary. By having a longer gearbox, the sheer torque the R10 has will be mitigated slightly and you'll use less fuel. If you find yourself getting loose, don't be afraid to mismatch tires. I've found I can get better tire wear on a Hard/Medium setup (Hards on front, Mediums on rear) because the extra grip on the rear forces me to drive easier on corner exits (given, this was before the 2.02 update, so whether this still applies is unknown).
And the only other tip I can give is to not force yourself. Twenty-four hours is a LONG time, and if you're not enjoying yourself, you'll find yourself making more and more mistakes. When I finally get around to doing this, I expect it to take me several weeks, setting aside an hour or two a day to do some, then save in a pit stop. The Tsukuba 9hr alone took me a full week to do in that way (I believe I was using the 908 as well, but that's another point).
