Can a car in GT5 take so much damage that it is no longer able to work

Well, if mechanical damage is enabled, it will be extremely frustrating and difficult to drive a car.
But no, you cannot total a car. Wheels don't come off

Pretty sure it's online only, and that's also adjustable between heavy and light (I think).

Arcade and practice modes can have mechanical damage as well
