what settings in general would you use to get that absolute fastest 400 you can?

it's been done many times, search for it.
the most basic thing you'd want to do is to set your transmission as low as possible, test it, and maybe go up 2 or 3.
lower the car a bit, more on one end depending on the drivetrain (FR - rear, FF - front, MR - even), and possibly adjust the camber. (after everything else)

Well for Maximum Acceleration you need to adjust the Gear Ratio, and the LSD...:smilewink

Aero-dynamically I'm not sure what's best for acceleration?

Lower it to all shoot... change your gear ratios to a closer setting (seeming how its only 400m or 1/4mile) TCS on 8 or 9 ACM doesnt really matter but 8 to be safe... and a bit more downforce.

more downforce creates more drag!

It all depends and the drivetrain i think but i always try to make the shocks as hard as i can for under the rotating wheels a least for the best traction
