After several people reported Rockstar being very picky about that's uploaded to Youtube, they've finally released guidelines on what they'll allow:

Essentially, you can't show the ending the game, nor can you post a youtube video of ONLY cutscenes, no making money off of the videos and no in-game entertainment. It sounds like Let's Plays are fine to show everything but the ending, now.
