I've run the Audi R10 in the Indy 500 on Racing Hard tires and won. It was a battle throughout, but I won. I changed the suspension settings and I don't have them at hand, but I know that I put full downforce on. Anything else and the back just got too twitchy too fast. A bump from a competitor, especially with worn tires, and the car would snap into a spin before I knew what happened (of course, I use the DS3, so that doesn't help.)
Anyway, I ran two 23 and seven 22 lap stints with the tires, and as long as you keep the car settled and don't do anything stupid to get yourself into trouble you'll be fine. My average lap was probably right at 44.000 with a fastest lap in the low 42s. Anything that'll do 9 stops and 44 flat should win it...you might even be able to with 10 stops.
I can't speak as to the fuel economy question, but chaosdd made a good point regarding that.
For an in-depth look at another of my races in the same car at Indy, check this thread. It was a loss, but largely through my own fault, and was extremely similar to my win in terms of my setup and lap times, etc. The only major differences were an increase in rear downforce and switching from 10 to 9 stops.
