This has likely been discussed before but I guess I missed it. What is the best way to change the gearing and diff and speed in GT5, I can't seem to get it right

A popular method is to set 1st gear all the way left and top gear all the way right, then split the difference with the middle gears (spaced evenly between the two ends) and then use final drive ratio to change the top speed. Setting the top speed first will change the ratio range among the individual gears.
 
I usually do test laps/races with the car and pay attention to the tach with each gear change and then adjust accordingly.

On an unrealted note, DDAAVVIIDD82 that has to be the single best signature I have ever read.
 
I find Excel to be a very useful tool for calculating theoretical top speed in each particular gear, and it's possible to determine the theoretical tire diameter (when an obvious tire size isn't available (such as an Enzo (345/35ZR19) versus a Toyota 88C-V (?))) through averaging driven speed in the same gear at different RPMs. Using Excel, and providing a driven tire diameter, I can determine perfect gear splits that allow for the best use of the engine's powerband. Visible gear splits on the sliding scale isn't always enough to satisfy me and when it isn't, I take great satisfaction in knowing I can satisfy myself using a different method. Doubly satisfied, win!
