Is that video really, really, really long, or is it uncompressed?
The only way you'll get people to download an uncompressed video of that size is if you torrent it or offer it up as an ordinary download (MegaUpload.com can do that) anyway, so you might as well take the time to compress it to something more reasonable.
If you like YouTube, they provide the specs for the "optimum" conversion into their format -- 320x240 MPEG4 compressed with DivX or Xvid, with MP3 audio and 30 frames per second.
If that's too "crappy," Veoh.com accepts larger, higher-quality files (I've seen a 300MB 25-minute one), but you're still going to want to compress it at least 50%.
